Traditional Kenyan herbal medicine: exploring natural products’ therapeutics against schistosomiasis

Praziquantel (PZQ) remains the only drug of choice for the treatment of schistosomiasis, caused by parasitic flatworms. The widespread use of PZQ in schistosomiasis endemic areas for about four decades raises concerns about the emergence of resistance of Schistosoma spp. to PZQ under drug selection pressure. This reinforces the urgency in finding alternative therapeutic options that could replace or complement PZQ. We explored the potential of medicinal plants commonly used by indigenes in Kenya for the treatment of various ailments including malaria, pneumonia, and diarrhoea for their antischistosomal properties. Employing the Soxhlet extraction method with different solvents, seven medicinal plants Artemisia annua, Ajuga remota, Bredilia micranta, Cordia africana, Physalis peruviana, Prunus africana and Senna didymobotrya were extracted. Qualitative phytochemical screening was performed to determine the presence of various phytochemicals in the plant extracts. Extracts were tested against Schistosoma mansoni newly transformed schistosomula (NTS) and adult worms and the schistosomicidal activity was determined by using the adenosine triphosphate quantitation assay. Phytochemical analysis of the extracts showed different classes of compounds such as alkaloids, tannins, terpenes, etc., in plant extracts active against S. mansoni worms. Seven extracts out of 22 resulted in <20% viability against NTS in 24 h at 100 μg/ml. Five of the extracts with inhibitory activity against NTS showed >69.7% and ≥72.4% reduction in viability against adult worms after exposure for 24 and 48 h, respectively. This study provides encouraging preliminary evidence that extracts of Kenyan medicinal plants deserve further study as potential alternative therapeutics that may form the basis for the development of the new treatments for schistosomiasis.

How effective are states at assessing and predicting the nuclear intentions of foreign countries? Drawing on close to 200 US assessments of foreign countries’ proliferation intentions between 1957 and 1966, this research note finds that close to 80 percent of testable US assessments were correct and that they shifted from highly inaccurate in the late 1950s to highly accurate in the 1960s. Based on quantitative and qualitative analysis, I conclude that learning from early failures led the intelligence community to achieve higher accuracy.

A national need is to prepare for and respond to accidental or intentional disasters categorized as chemical, biological, radiological, nuclear, or explosive (CBRNE). These incidents require specific subject-matter expertise, yet have commonalities. We identify 7 core elements comprising CBRNE science that require integration for effective preparedness planning and public health and medical response and recovery. These core elements are (1) basic and clinical sciences, (2) modeling and systems management, (3) planning, (4) response and incident management, (5) recovery and resilience, (6) lessons learned, and (7) continuous improvement. A key feature is the ability of relevant subject matter experts to integrate information into response operations. We propose the CBRNE medical operations science support expert as a professional who (1) understands that CBRNE incidents require an integrated systems approach, (2) understands the key functions and contributions of CBRNE science practitioners, (3) helps direct strategic and tactical CBRNE planning and responses through first-hand experience, and (4) provides advice to senior decision-makers managing response activities. Recognition of both CBRNE science as a distinct competency and the establishment of the CBRNE medical operations science support expert informs the public of the enormous progress made, broadcasts opportunities for new talent, and enhances the sophistication and analytic expertise of senior managers planning for and responding to CBRNE incidents.
Passive range of motion exercise to enhance growth in infants following the Norwood procedure: a safety and feasibility trial

Objective
The aim of this study was to evaluate the safety and feasibility of a passive range of motion exercise programme for infants with CHD.
Study designThis non-randomised pilot study enrolled 20 neonates following Stage I palliation for single-ventricle physiology. Trained physical therapists administered standardised 15–20-minute passive range of motion protocol, for up to 21 days or until hospital discharge. Safety assessments included vital signs measured before, during, and after the exercise as well as adverse events recorded through the pre-Stage II follow-up. Feasibility was determined by the percent of days that >75% of the passive range of motion protocol was completed.
ResultsA total of 20 infants were enrolled (70% males) for the present study. The median age at enrolment was 8 days (with a range from 5 to 23), with a median start of intervention at postoperative day 4 (with a range from 2 to 12). The median hospital length of stay following surgery was 15 days (with a range from 9 to 131), with an average of 13.4 (with a range from 3 to 21) in-hospital days per patient. Completion of >75% of the protocol was achieved on 88% of eligible days. Of 11 adverse events reported in six patients, 10 were expected with one determined to be possibly related to the study intervention. There were no clinically significant changes in vital signs. At pre-Stage II follow-up, weight-for-age z-score (−0.84±1.20) and length-for-age z-score (−0.83±1.31) were higher compared with historical controls from two earlier trials.
ConclusionA passive range of motion exercise programme is safe and feasible in infants with single-ventricle physiology. Larger studies are needed to determine the optimal duration of passive range of motion and its effect on somatic growth.

Approximately half of the variation in wellbeing measures overlaps with variation in personality traits. Studies of non-human primate pedigrees and human twins suggest that this is due to common genetic influences. We tested whether personality polygenic scores for the NEO Five-Factor Inventory (NEO-FFI) domains and for item response theory (IRT) derived extraversion and neuroticism scores predict variance in wellbeing measures. Polygenic scores were based on published genome-wide association (GWA) results in over 17,000 individuals for the NEO-FFI and in over 63,000 for the IRT extraversion and neuroticism traits. The NEO-FFI polygenic scores were used to predict life satisfaction in 7 cohorts, positive affect in 12 cohorts, and general wellbeing in 1 cohort (maximal N = 46,508). Meta-analysis of these results showed no significant association between NEO-FFI personality polygenic scores and the wellbeing measures. IRT extraversion and neuroticism polygenic scores were used to predict life satisfaction and positive affect in almost 37,000 individuals from UK Biobank. Significant positive associations (effect sizes <0.05%) were observed between the extraversion polygenic score and wellbeing measures, and a negative association was observed between the polygenic neuroticism score and life satisfaction. Furthermore, using GWA data, genetic correlations of -0.49 and -0.55 were estimated between neuroticism with life satisfaction and positive affect, respectively. The moderate genetic correlation between neuroticism and wellbeing is in line with twin research showing that genetic influences on wellbeing are also shared with other independent personality domains.

Building on the rationalist literature on sanctions, this article argues that economic and political sanctions are a successful tool of nonproliferation policy, but that selection effects have rendered this success largely hidden. Since the late 1970s—when the United States made the threat of sanctions credible through congressional legislation and began regularly employing sanctions against proliferating states—sanctions have been ineffective in halting ongoing nuclear weapons programs, but they have succeeded in deterring states from starting nuclear weapons programs in the first place and have thus contributed to a decline in the rate of nuclear pursuit. The logic of the argument is simple: rational leaders assess the risk of sanctions before initiating a nuclear weapons program, which produces a selection effect whereby states highly vulnerable to sanctions are deterred from starting nuclear weapons programs in the first place, so long as the threat is credible. Vulnerability is a function of a state's level of economic and security dependence on the United States—states with greater dependence have more to lose from US sanctions and are more likely to be sensitive to US-sponsored norms. The end result of this selection effect is that since the late 1970s, only insulated, inward-looking regimes have pursued nuclear weapons and become the target of imposed sanctions, thus rendering the observed success rate of nonproliferation sanctions low. I find support for the argument based on statistical analysis of a global sample of countries from 1950 to 2000, an original data set of US nonproliferation sanctions episodes, and qualitative analysis of the South Korean and Taiwanese nuclear weapons programs.
Political Devolution and Resistance to Foreign Rule: A Natural Experiment

Do foreign occupiers face less resistance when they increase the level of native governing authority? Although this is a central question within the literature on foreign occupation and insurgency, it is difficult to answer because the relationship between resistance and political devolution is typically endogenous. To address this issue, we identify a natural experiment based on the locally arbitrary assignment of French municipalities into German or Vichy-governed zones during World War II. Using a regression discontinuity design, we conclude that devolving governing authority significantly lowered levels of resistance. We argue that this effect is driven by a process of political cooptation: domestic groups that were granted governing authority were less likely to engage in resistance activity, while violent resistance was heightened in regions dominated by groups excluded from the governing regime. This finding stands in contrast to work that primarily emphasizes structural factors or nationalist motivations for resistance.

Observation of Long-Range Orientational Ordering in Metal Films Evaporated at Oblique Incidence onto Glass

We studied long-range orientational ordering in polycrystalline Au films (10 nm - 30 nm) that are evaporated at oblique incidence onto a glass substrate at room temperature. By measuring the averaged optical second-harmonic response from the films over a 6-mm diameter region, we observed a transition from the expected in-plane mirror symmetry at 10 nm to a surprising three-fold in-plane rotational symmetry at 30 nm. X-ray pole figure analysis performed on these films showed the strong <111> fiber texture typical of fcc films, but with a restricted, three-fold symmetric, distribution of crystallite orientations about the fiber axis.
